Anavex (NASDAQ: AVXL) wins FDA clearance for Fragile X study and opens IND

What happened

Anavex Life Sciences Corp. (NASDAQ: AVXL) said the FDA notified it that the company may proceed with its planned Fragile X study.

The IND for ANAVEX 2-73, also called blarcamesine, is now open in the United States.

Anavex said it now has open INDs in Alzheimer's disease, Rett syndrome and Fragile X syndrome.

The planned Phase 2a study will enroll approximately 60 adult males and will assess safety, tolerability and pharmacokinetics.

The study is randomized, double-blind, placebo-controlled and dose-ranging.

Secondary endpoints include the Aberrant Behavior Checklist revised for FXS and the NIH Toolbox.

The company also said blarcamesine has Orphan Drug Designation from the FDA for Fragile X syndrome.

Anavex also said newly published peer-reviewed data in Frontiers in Pharmacology strengthen the scientific rationale for the program.

Why it matters

OptimistFi's case is that AVXL needs regulatory-grade validation of its precision-medicine approach before cash burn and dilution erode the upside.

This filing strengthens that case by opening the Fragile X IND and moving the program into planned human testing.

The release points to a September 2026 Frontiers in Pharmacology paper that reported 14 days of oral blarcamesine produced dose-related improvement in EEG abnormalities in a mouse model.

That is preclinical support, not a human efficacy result.

The release says no FDA-approved treatment currently exists for Fragile X syndrome.

It estimates about 38,000 to 87,000 affected individuals in the United States and about 777,000 to 1.4 million worldwide.

Those figures define the patient pool, and the next step is human data from the planned study.

The company described Fragile X syndrome as the most common inherited cause of intellectual disability and developmental delay, and the most commonly known single-gene cause of autism.

What's next

Next comes ANAVEX2-73-FXS-001, the planned Phase 2a randomized, double-blind, placebo-controlled, dose-ranging study in approximately 60 adult males with FXS.

Anavex said behavioral measures from the Aberrant Behavior Checklist revised for FXS and cognitive measures from NIH Toolbox will be secondary endpoints.

With an open IND, Anavex said it is positioned to advance ANAVEX2-73-FXS-001 in the United States.

A clean safety and tolerability read would help the validation thesis, while a poor tolerability signal would hurt it.
